ABSTRACT

The objective was to study the effectiveness of the growth regulator (ANA + GA3) associated or not to the application of adjuvant and artificial pollination in 'Gefner' atemoya. The experiment was conducted in the experimental orchard at Florida's Tropical Research and Education Center. The experimental design was in a randomized block, with 14 treatments, 10 repetitions and 3 flowers per plot. The highest percentages of fixed fruits were obtained with hand pollination ­ HP and 450 NAA + 1250 GA3 mg L-1 + adjuvant and HP. The use of hand pollination for 'Gefner' atemoya tree proved to be the most efficient method so far. Applying growth regulator without artificial pollination produces parthenocarpic fruits, however with high rate of abortions, and small fruits. Growth regulators together with hand pollination produces small and uneven fruits, and cause reduction in the fruits' titratable acidity. The use of adjuvant caused low fixation and toxicity to fruits, and its use is not recommended.

ABSTRACT

Aims: Ocimum gratissimum is an aromatic and medicinal plant, well known for its medicinal values such as antifungal properties. This study aimed at evaluating the effect of T. diversifolia powder and compost use as biofertilizer on the growth parameters, essential oil (EO), total phenolic and flavonoid content and anticandida activity of O. gratissimum. Study Design: O. gratissimum plants were cultivated for 8 months in an experimental farm designed as a split plot into 4 blocks. Each block was amended (main factor) either with T. diversifolia compost (150 g/plant), powder (40 g/plant) or the synthetic fertilizer NPK (10 g/plant), respectively, followed by sprayed (second factor) with same fertilizer at 20 g/L, 20 g/L and 2 g/L or water every two weeks after transplantation. The control block received no amendment and was sprayed with water or the previous fertilizer. Place and Duration of Study: This work was carried out within August 2019 to October 2020 in Yaoundé-Cameroon. Methodology: Plant growth parameters (fresh and dry leaves weight, florescent weight and plant height) were evaluated at four and eight months after transplantation. The harvested fresh leaves were hydrodistillated for EO and the hot aqueous extract. Both extracts were used for the evaluation of the anticandida activity while the latter was submitted to total flavonoids and phenolic analyses. Results: At 4 and 8 months after transplantation, the synthetic fertilizer and T. diversifolia compost significantly increased plant growth parameters as compared to other treatments. The plants treated with T. diversifolia biofertilizer compost showed the highest total phenolic (53.16 µg GAE/µL), flavonoid (36.32 µg// GAE/µL) content, and EO yield (0.666%). The EO from O. gratissimum treated with T. diversifolia compost showed the best inhibitory activity on C. albicans NR-29451. Conclusion: This study showed that T. diversifolia compost was a promising organic fertilizer in optimizing the growth, secondary metabolites and anticandida activity of O. gratissimum.

ABSTRACT

The objective of this work was to carry out the in vitro establishment of Echynochloa polystachya aiming at obtaining a micropropagation protocol for works involving the selection of superior genotypes and the cultivation of the species. E. polystachya stems were collected in the municipality of Manaus-AM. Explants were inoculated in test tubes containing Murashige and Skoog (MS) medium. Thirty days after in vitro establishment, the rate of sprouting and contamination were evaluated. Experiments were also carried out to assess the effects of sucrose and 6-benzylaminopurine (BAP) concentrations on the tillering rate of explants. It was found that during the successive subcultures there was a decrease in internodes and the consequent loss of vigor. There were responses in the multiplication rate at concentrations starting from 45 g L-1 sucrose. In addition, BAP and sucrose interfered the development and in vitro multiplication. Sucrose in conjunction with BAP was harmful and shortened internodes. The physiological state of the explants for the species under study was intrinsically linked to the concentrations of sucrose used for the culture medium and the concentrations of BAP. However, the sucrose and BAP concentrations suggested for in vitro cultivation of E. Polystachya must be adjusted during successive subcultures. Absence of contamination in the in vitro establishment occurred at concentrations 15, 30 and 60 g L-1 sucrose. The combination of 1.5 mg L-1 BAP and 30 g L-1 sucrose promoted greater induction of sprouts. In addition, the in vitro rooting of E. polystachya was 45%.

ABSTRACT

As a result of biodiversity and ecosystem service losses associated with biological invasions, there has been growing interest in basic and applied research on invasive species aiming to improve management strategies. Tradescantia zebrine is a herbaceous species increasingly reported as invasive in the understory of disturbed forest ecosystems. In this study, we assess the effect of spatial and seasonal variation on biological attributes of this species in the Atlantic Forest. To this end, we measured attributes of T. zebrina associated with plant growth and stress in the four seasons at the forest edge and in the forest interior of invaded sites in the Iguaçu National Park, Southern Brazil. The invasive plant had higher growth at the forest edge than in the forest interior and lower leaf asymmetry and herbivory in the winter than in the summer. Our findings suggest that the forest edge environment favours the growth of T. zebrina. This invasive species is highly competitive in the understory of semi-deciduous seasonal forests all over the year. Our study contributes to the management of T. zebrina by showing that the summer is the best season for controlling this species.


As perdas de biodiversidade e os seus serviços ecossistêmicos ocasionadas pelas invasões biológicas, têm despertado o interesse em pesquisas básicas e aplicadas sobre as espécies invasoras com o objetivo de buscar estratégias de manejo. Tradescantia zebrina é uma das herbáceas crescentemente relatadas como uma invasora no sub-bosque florestal de ecossistemas impactados. Neste estudo, nós estudamos o efeito da variação espacial e sazonal sobre atributos biológicos desta espécie na Floresta Atlântica. Assim, nós mensuramos atributos de T. zebrina associados com o crescimento vegetal e o estresse nas quatro estações do ano em borda e interior da floresta de locais invadidos no Parque Nacional do Iguaçu, Sul do Brasil. A planta invasora apresentou maior crescimento na borda florestal do que no interior. Adicionalmente, assimetria foliar e herbivoria obtidos no inverno foram menores quando comparados ao verão. Nossos achados sugerem que os ambientes de borda florestal favorecem o crescimento de T. zebrina. Esta espécie invasora é altamente competitiva no sub-bosque da Floresta Estacional Semidecídua durante o ano todo. Nosso estudo contribui com o manejo de T. zebrina mostrando que o verão é a melhor estação para o controle desta espécie.

Endophytic bacteria Bacillus safensis RS95 and Pseudomonas hibiscicola RS121 were evaluated for their ability to promote the growth of rice seedlings and produce indole-acetic acid (IAA) and siderophores and to solubilize phosphates. 'Guri' rice seeds were immersed in bacterial endophyte cell suspensions (separated and two-strain mixed), as well as in Escherichia coli DH5α, phosphate-buffered saline (PBS) and water treatments (negative controls). Seeds were sown on agar-water in Petri plates placed vertically at an angle of 65°. The ability of plant growth-promoting endophytic bacteria (PGPEB) to produce IAA and siderophores was determined by Salkowski colorimetric and chrome azurol S (CAS) assays, respectively. Mineral phosphate solubilization activity was calculated by inoculating the endophytes onto medium containing insoluble phosphate. PGPEB showed a positive effect on the growth of rice seedlings, causing a mean growth of shoots and primary-roots of 60 and 67%, respectively. Bacterial strains also showed positive traits for IAA and siderophore production, as well as phosphate-solubilization activity

ABSTRACT

Aims@#The objective of this study was to analyze the genome of endophytic actinomycete associated with orchids and evaluate its plant hormone activities, including phytohormone, siderophore, ammonia production, zinc and phosphate solubilization.@*Methodology and results@#Strain DR1-2 isolated from the roots of the Thai orchid, Dendrobium christyanum Rchb.f., was closely related to Pseudonocardia alni DSM 44104T, P. antarctica DSM 44749T and P. carboxydivorans Y8T (99.93-100% similarity) based 16S rRNA gene sequence. This strain exhibited IAA production (294.10 ± 12.17 μg/mL), phosphate solubilization (2.20 ± 0.08 solubilization Index, SI), positive for siderophore production and ammonia production (36.99 ± 2.24 μg/mL). It showed a maximum IAA of 489.73 ± 8.90 μg/mL, when optimized using 0.5% Ltryptophan, pH 6 and incubated at 30 °C for 7 days. The IAA of strain enhanced the root length, shoot length, number of roots and fresh weight of rice seedlings (Oryza sativa L. cv. RD49). The draft genome of strain DR1-2 was 6,077,423 bp in 23 contigs with G+C content of 74.6%. The average nucleotide identity-Blast (ANIb) and average nucleotide identity-MUMmer (ANIm) values of strain DR1-2 and related type strains were 95.81 to 97.25% and the digital DNA-DNA hybridization (dDDH) values were 72.60 to 74.00%, respectively. Genomic analysis of strain DR1-2 revealed that the gene encodes the enzyme involved in the phytohormones biosynthesis and gene clusters involved in the biosynthesis of bioactive metabolites.@*Conclusion, significance and impact of study@#Endophytic actinomycete, Pseudonocardia strain DR1-2 from Thai orchid, D. christyanum Rchb.f., exhibited significant IAA production and affected the growth of the plant, which was the potential source of plant hormones for agricultural applications.

This study goal to evaluate the effects of different concentrations of lead (Pb) and silver (Ag) on germination, initial growth and anatomical alterations of Lactuca sativa L. Plants use various mechanisms to reduce the impacts caused by anthropic action, such as xenobiotic elements of soils and water contaminated by heavy metals. These metals were supplied as lead nitrate and silver nitrate and the following treatments were established: control for both metals, maximum dose of heavy metals, for arable soils, allowed by the National Council of the Environment (Ag = 25 mg. Kg-1, Pb = 180 mg. Kg-1), double (Ag = 50 mg. Kg-1, Pb = 360 mg. Kg-1) and triple (Ag = 75 mg. Kg-1, Pb = 540 mg. Kg -1) of this dosage. Vigor and germination tests of the seeds and possible anatomical changes in the leaves and roots of lettuce plants were performed. The species showed a high capacity to germinate under Pb and Ag stress, and the germination was never completely inhibited; however, the germination decreased with increasing Pb concentrations, but not under Ag stress. The use of increasing doses of metals reduced seed vigor and increased chlorophyll content. An increase in biomass was also observed in plants from treatments submitted to Pb. The phytotoxic effects of metals were more pronounced at 15 days after sowing. Anatomically, L. sativa was influenced by metal concentrations, and had a reduction of up to 79.9% in root epidermis thickness at the highest Pb concentration, although some structures did not suffer significant changes. The results suggest that L. sativa presents tolerance to high concentrations of heavy metals, showing possible mechanisms to overcome the stress caused by these metals. In this research lettuce possibly used the mechanism of exclusion of metals retaining Pb and Ag in the roots preserving the photosynthetic apparatus in the aerial part of the plants. In general, the chemical element Pb was more toxic than Ag, in these experimental conditions.


Este estudo teve como objetivo avaliar os efeitos de diferentes concentrações de chumbo (Pb) e prata (Ag) na germinação, crescimento inicial e alterações anatômicas de Lactuca sativa L. As plantas utilizam vários mecanismos para reduzir os impactos causados pela ação antrópica, como elementos xenobióticos de solos e água contaminada por metais pesados. Esses metais foram fornecidos como nitrato de chumbo e nitrato de prata e foram estabelecidos os seguintes tratamentos: controle para ambos os metais, dose máxima de metais pesados, para solos cultiváveis, permitida pelo Conselho Nacional do Meio Ambiente (Ag = 25mg.Kg-1, Pb = 180mg.Kg-1), dobro (Ag = 50mg.Kg-1, Pb = 360mg.Kg-1) e triplo (Ag = 75mg.Kg-1, Pb = 540 mg.Kg -1) desta dosagem. Foram realizados testes de vigor e germinação das sementes e possíveis alterações anatômicas nas folhas e raízes das plantas de alface. A espécie apresentou alta capacidade de germinar sob estresse de Ag e Pb, e a germinação nunca foi completamente inibida; entretanto, a germinação diminuiu com o aumento das concentrações de Pb, mas não sob estresse de Ag. O uso de doses crescentes dos metais, reduziu o vigor das sementes e aumentou o teor de clorofila. Também foi observado aumento da biomassa nas plantas a partir dos tratamentos submetidos ao Pb. Os efeitos fitotóxicos dos metais foram mais acentuados aos 15 dias após a semeadura. Anatomicamente, L. sativa foi influenciada pelas concentrações de metais, e teve uma redução de até 79,9% na espessura da epiderme radicular na maior concentração de Pb, embora algumas estruturas não tenham sofrido alterações significativas. Os resultados sugerem que L. sativa apresenta tolerância a altas concentrações de metais pesados, mostrando possíveis mecanismos para superar o estresse causado por esses metais. Nesta pesquisa a alface possivelmente utilizou o mecanismo de exclusão de metais retendo Pb e Ag nas raízes preservando o aparato fotossintético na parte aérea das plantas. De forma geral o elemento químico Pb se mostrou mais tóxico que Ag, nestas condições experimentais.

The results suggest that L. sativa presents tolerance to high concentrations of heavy metals, showing possible mechanisms to overcome the stress caused by these metals. In this research lettuce possibly used the mechanism of exclusion of metals retaining Pb and Ag in the roots preserving the photosynthetic apparatus in the aerial part of the plants. In general, the chemical element Pb was more toxic than Ag, in these experimental conditions.


Resumo Este estudo teve como objetivo avaliar os efeitos de diferentes concentrações de chumbo (Pb) e prata (Ag) na germinação, crescimento inicial e alterações anatômicas de Lactuca sativa L. As plantas utilizam vários mecanismos para reduzir os impactos causados pela ação antrópica, como elementos xenobióticos de solos e água contaminada por metais pesados. Esses metais foram fornecidos como nitrato de chumbo e nitrato de prata e foram estabelecidos os seguintes tratamentos: controle para ambos os metais, dose máxima de metais pesados, para solos cultiváveis, permitida pelo Conselho Nacional do Meio Ambiente (Ag = 25 mg.Kg-1, Pb = 180 mg.Kg-1), dobro (Ag = 50 mg.Kg-1, Pb = 360 mg.Kg-1) e triplo (Ag = 75 mg.Kg-1, Pb = 540 mg.Kg -1) desta dosagem. Foram realizados testes de vigor e germinação das sementes e possíveis alterações anatômicas nas folhas e raízes das plantas de alface. A espécie apresentou alta capacidade de germinar sob estresse de Ag e Pb, e a germinação nunca foi completamente inibida; entretanto, a germinação diminuiu com o aumento das concentrações de Pb, mas não sob estresse de Ag. O uso de doses crescentes dos metais, reduziu o vigor das sementes e aumentou o teor de clorofila. Também foi observado aumento da biomassa nas plantas a partir dos tratamentos submetidos ao Pb. Os efeitos fitotóxicos dos metais foram mais acentuados aos 15 dias após a semeadura. Anatomicamente, L. sativa foi influenciada pelas concentrações de metais, e teve uma redução de até 79,9% na espessura da epiderme radicular na maior concentração de Pb, embora algumas estruturas não tenham sofrido alterações significativas. Os resultados sugerem que L. sativa apresenta tolerância a altas concentrações de metais pesados, mostrando possíveis mecanismos para superar o estresse causado por esses metais. Nesta pesquisa a alface possivelmente utilizou o mecanismo de exclusão de metais retendo Pb e Ag nas raízes preservando o aparato fotossintético na parte aérea das plantas. De forma geral o elemento químico Pb se mostrou mais tóxico que Ag, nestas condições experimentais.

Aims@#Plant growth-promoting bacteria are the key components of a biofertilizer. This study was aimed to isolate and identify the predominant bacteria found in a foliar biofertilizer and characterizes the potential of the bacterial isolates as plant growth promoters.@*Methodology and results@#Potential bacteria with plant growth-promoting activities were isolated from a foliar biofertilizer on HiCrome™ Bacillus agar and Nutrient agar. Bacteria with unique colonial morphology were selected and categorized by Gram’s differential staining. Subsequently, the bacterial isolates were being further characterized for plant growth-promoting potentials, such as the production of indole acetic acid (IAA), 1-aminocyclopropane-1-carboxylate (ACC) deaminase and siderophore; as well as the ability of nitrogen fixation and phosphate/potassium solubilization. Based on the characterized traits, three bacterial isolates, namely M17, M22 and M52 showed great potential for being a plant growth promoter. Based on their 16S rRNA gene sequence analysis, M17, M22 and M52 were identified as Leclercia adecarboxylata, Margalitia shackletonii and Lysinibacillus pakistanensis, respectively.@*Conclusion, significance and impact of study@#Bacterial isolates exhibiting plant growth-promoting activities were successfully isolated from a biofertilizer and identified in this study. This finding provides an insight into the potential bacteria of a foliar fertilizer that may promote plant growth. Identification of these plant-growth promoters may help the scientists and agrochemical manufacturers to determine and disclose the key microorganisms of their biofertilizers, thereby contributing to the improvement of biofertilizers and promoting them as reliable alternatives to chemical fertilizers.

ABSTRACT

Aims@#Endophytic bacteria (EB) living inside plant tissues possess different beneficial traits including siderophore production and other plant growth-promoting (PGP) activities. Siderophore-producing EB promote host plant growth by secreting ferrum in iron-deficient conditions. This study screened 19 siderophore producers in vitro, isolated from upland rice roots grown in mountain farms of Tung Village, Nậm Có Commune, Mù Cang Chải District, Yên Bái Province, Vietnam, for PGP traits, including phosphate solubilisation, indole-3-acetic acid (IAA), ammonia, gelatinase, amylase and catalase production.@*Methodology and results@#The bacteria were identified by Matrix assisted Laser Desorption Ionization Time of Flight mass spectrometry (MALDI-TOF MS). All 19 isolates were identified as genera Pseudomonas, Enterobacter, Pantoe, Bacillus, Burkholderia, Staphylococcus, Ralstonia and Cronotacter. The isolates produced catalase and ammonia. The amount of ammonia ranged from 60.74 ± 0.14 to 466.72 ± 0.18 mg/L. Out of the 19 siderophore producers, 17 (89.47%) were able to solubilise phosphate with solubilisation index (PSI) ranging from 1.12 ± 0.07 to 2.14 ± 0.15. The qualitative assays identified 12 isolates (63.15%) positive for IAA production with a tryptophan concentration of 5 mM, whereas 15 (78.94%) and 17 (89.47%) isolates were positive for gelatin and starch hydrolysis, respectively. Especially, 7 isolates were found to be positive for all tested assays in vitro including Pseudomonas rhodesiae (NC2), Enterobacter asburiae (NC50), Pantoea ananatis (NC63), Bacillus cereus (NC64), Burkholderia cenocepacia (NC110), Staphylococcus sciuri (NC112) and Ralstonia pickettii (NC122).@*Conclusion, significance and impact of study@#This study serves as crucial findings of multi-trait plant growth-promoting endophytic bacteria isolated from upland rice root in north-western Vietnam. The seven potential isolates positive for all tested assays could be effective PGP bacteria for bio-inoculants.

ABSTRACT

ObjectiveTo study the effects of different plant growth-promoting rhizobacteria (PGPR) on the growth of Paris polyphylla var. yunnanensis seedlings and the quality of its medicinal parts, in order to provide reference for the cultivation of high-quality P. polyphylla var. yunnanensis. MethodThe pot culture experiment at room temperature and the single-factor completely random design were employed for exploring the effects of five PGPR on physiological characteristics and inorganic elements of P. polyphylla var. yunnanensis. ResultThe results showed that the exogenous inoculation of different PGPR promoted the growth and development of P. polyphylla var. yunnanensis to varying degrees, delayed the senescence of leaves, and improved the medicinal value of new and old rhizomes. Compared with the non-inoculated control, the exogenous inoculation of compound microbial fertilizer (FH) and microbial agent Sanju Guanjin liquid (SJ) enhanced the root vigor, increased the content of photosynthetic pigments and the activities of anti-oxidant enzymes [superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD)], and reduced the content of malondialdehyde (MDA) in leaves. Their inhibition rates against MDA were 10.46%-39.62% and 20.99%-53.12%, respectively. With the growth of P. polyphylla var. yunnanensis, the inhibition rate against MDA gradually increased, which effectively delayed the senescence of P. polyphylla var. yunnanensis leaves. In addition, the exogenous inoculation of different PGPR promoted the accumulation of nutrient elements in new and old rhizomes, lowered the heavy metal content to varying degrees, and improved the medicinal value of P. polyphylla var. yunnanensis rhizomes. ConclusionFH and SJ have exhibited the best promoting effect on the growth of P. polyphylla var. yunnanensis seedlings and also the best regulatory effect on the medicinal value of P. polyphylla var. yunnanensis rhizomes, which has provided reference for the application and promotion of PGPR in the growth of P. polyphylla var. yunnanensis.

ABSTRACT

Plant growth regulator (PGR) is mostly a class of chemical synthesis substance with physiological activities similar to plant hormones,which can promote cell elongation,induce vascular differentiation or accelerate tissue aging via regulating the physiological processes such as photosynthesis,respiration,transpiration,signal transduction,substance absorption and operation. PGR has the advantages of small dosage,high efficiency,low toxicity and less residue,and it is widely used in the planting of Chinese medicinal herbs. By consulting the relevant literature published in recent years,this paper briefly summarizes the main types of PGR,e.g.auxins,gibberellins,cytokinins,abscisic acid and ethylene,etc. On the other hand,this article analyzes and sums up the specific applications of PGR in the manufacture of Chinese herbal medicine,for instance,promoting seed germination,improving seed setting rate or fruit setting rate,dwarfing plants,inhibiting reproductive growth,regulating gender differentiation,stimulating fruit falling,enhancing resistance and so on. The problems existing in the practical use of PGR are pointed out,non-differentiation of specific species,unreasonable combination,not paying attention to the operation method,arbitrarily increasing the dose,lack of residue limit standard and reducing the content of some effective components,for example.Meanwhile,some feasible suggestions are put forward.Not only the suitable types of PGR should be selected in a reasonable and standardized manner,but also the appropriate concentration,dosage and period of application should be chosen carefully; the dual effects of PGR on plant growth and active ingredients in medicinal organs should be concerned,so as to improve the yield and avoid the loss of effective components on the basis of ensuring the quality of Chinese medicinal materials; it is necessary to strengthen the registration of PGR in the production of Chinese medicinal materials and establish residue limit standards to provide a monitoring basis for ensuring the safety of Chinese medicine in the future.The scientific use of PGR can promote the increase of agricultural yield and farmers' income,and make the healthy development of Chinese herbal medicine planting industry.

ABSTRACT

ObjectiveTo study the effects of foliar spraying of two kinds of compound rhizosphere growth-promoting agents on the growth and physiological characteristics of Angelicae Sinensis Radix (ASR), as well as the pharmacodynamic components, in order to lay a foundation for providing functional microbial agents for ecological cultivation of ASR. MethodThe compound growth-promoting agents T1 (Pseudomonas CBS5, CBS7 and CBSB) and T2 (Bacillus 5C1, 5C5 and 5C7) with the concentration of 1×108 CFU·mL-1 were sprayed on the leaf surface of the field, and the sterile potato glucose broth medium was used as the control (CK). The plant growth indexes of ASR were measured by conventional methods, the photosynthetic physiological indexes of ASR were measured by portable photosynthetic measurement system, the enzyme activities of plants and microorganisms were measured by kit method, and the endogenous hormone levels were analyzed by ultra-performance liquid chromatography tandem mass spectrometry. The contents of ferulic acid, senkyunolide I, coniferyl ferulate, senkyunolide A and Z-ligustilide were determined by high performance liquid chromatography. ResultCompared with CK, the two compound inoculants could promote the growth of ASR and increase the biomass, increase the leaf net photosynthetic rate, stomatal conductance, intercellular CO2 concentration, transpiration rate, increase catalase, peroxidase, superoxide dismutase, polyamine oxidase, diamine oxidase and polyphenol oxidase enzyme activities, increase endogenous jasmonic acid, cytokinin and gibberellin levels in plants, increase the contents of ferulic acid, senkyunolide A and Z-ligustilide, reduce the contents of malondialdehyde and abscisic acid, and reduce the incidence of root rot. ConclusionFoliar spraying of two kinds of rhizosphere compound growth-promoting agents can promote the growth, photosynthesis and stress resistance of ASR, and can improve the quality of ASR in different degrees. Comprehensive analysis shows that T1 treatment is better than T2 treatment in the growth-promoting and quality-enhancing of ASR.

In this study, the effects of two plant growth-promoting bacteria Klebsiella michiganensis TS8 and Lelliottia Jeotgali MR2 on the growth and cadmium (Cd) uptake of Arabidopsis thaliana under Cd stress were explored. A wild-type Arabidopsis thaliana was selected as the experimental plant and was planted at different Cd concentrations. MR2 and TS8 bacterial suspensions were sprayed onto the rhizospheric soil during the planting process. The initial Cd concentration of the bought soil was 14.17 mg/kg, which was used as the pot soil of the low-concentration Cd treatment group (LC). The concentration of soil Cd at high-concentration Cd treatment group (HC) were 200 mg/kg higher than that at LC group. Compared with the control group, MR2 suspension significantly promoted the growth of A. thaliana at both low and high concentrations, while TS8 strain and MR2_TS8 mixture only exhibited growth-promoting effect at high concentration. However, it was noteworthy that, TS8 suspension significantly reduced the Cd content in the underground parts of A. thaliana (60% and 59%), and significantly improved the Cd content in the aboveground parts of A. thaliana (234% and 35%) at both low and high concentrations. In addition, at low concentration, both single strain and mixed strains significantly improved the transformation from reducible Cd to acid-extractable Cd in soil, promoted Cd intake, and thereby reduced the total Cd content in soil. Therefore, the rational application of plant growth-promoting bacteria may improve crop yield and remediate Cd contamination in soil.

RESUMEN Las rizobacterias forman parte de la gran cantidad de microorganismos que actúan como agentes de biocontrol, produciendo metabolitos que inducen resistencia sistémica en las plantas que inhiben el crecimiento de patógenos. El objetivo de esta investigación fue evaluar la capacidad de diez rizobacterias de los géneros Rhizobium, Bradyrhizobium, Sinorhizobium, Ochrobactrum y Pseudomonas para producir ácido cianhídrico (HCN), sideróforos y ácido indol-acético (AIA), disolver fosfato, fijar nitrógeno e inhibir el crecimiento de fitopatógenos. Se realizaron todas las pruebas fisiológicas y bioquímicas correspondientes, así como la prueba de antagonismo in vitro contra los fitopatógenos Fusarium oxysporum, Colletotrichum gloeosporioides y Rhizoctonia solani. Cinco cepas produjeron una mayor cantidad de AIA en relación a las otras en presencia de triptófano, la cepa ES1 (Ochrobactrum sp.) produjo HCN, el 50 % de las cepas evaluadas liberaron sideróforos, el 60 % disolvió fósforo, y todas resultaron positivas para la fijación de nitrógeno. Nueve cepas inhibieron el crecimiento de F. oxysporum entre 40 % y 65 %, la cepa Alf (Pseudomonas fluorescens) inhibió además el crecimiento de C. gloeosporioides en un 22 %, y ninguna inhibió el crecimiento de R. solani. Los rizobios evaluados y la cepa de Pseudomonas fluorescens podrían ejercer efectos beneficiosos sobre las plantas a través de mecanismos directos e indirectos, o una combinación de ambos, lo que las convierte en una opción sostenible para la producción de cultivos.


ABSTRACT Rhizobacteria are part of the large number of microorganisms that act as biocontrol agents, producing metabolites that induce systemic resistance in plants and inhibit the growth of pathogens. The objective of this research was to evaluate the capacity of ten rhizobacteria of the genera Rhizobium, Bradyrhizobium, Sinorhizobium, Ochrobactrum and Pseudomonas to produce hydrogen cyanide (HCN), siderophores and indole acetic acid (IAA), dissolve phosphate, fix nitrogen and inhibit the growth of phytopathogens. All the corresponding physiological and biochemical tests were carried out, in addition to an in vitro antagonism test against the phytopathogens Fusarium oxysporum, Colletotrichum gloeosporioides and Rhizoctonia solani. Five strains produced a greater amount of IAA with respect to the others in the presence of tryptophan, the strain ES1 (Ochrobactrum sp.) produced HCN, 50% of the evaluated strains released siderophores, 60% solubilized phosphorus and all were positive for nitrogen fixation. Nine strains inhibited the growth of F. oxysporum by 40% to 65%. The Alf strain (Pseudomonas fluorescens) inhibited the growth of C. gloeosporioides by 22% while none inhibited the growth of R. solani. The rhizobia tested and the Pseudomonas fluorescens strain may have favorable effects on plants through direct and indirect mechanisms, or a combination of both, making them a sustainable option for crop production.

Worldwide, Brazil holds the fifth position in melon fruits exportation, further expanding its products to provide for the growing demand. This expansion is the result of the development and application of new technologies, including the management of the use of biostimulants. However, for melon crops, the information in the literature on the use of biostimulants remains limited to the effects of different doses on fruit quality at the time of harvest. Therefore, this study aimed to evaluate the influence of different methods of pre-harvest application of two biostimulants on the production and postharvest conservation of fruits of yellow melon cv. Iracema. The treatments consisted of a combination of three factors: two plant biostimulants (Crop Set® and Spray Dunger®), two application methods of the products (fertigation and spraying), and five times of postharvest storage (0, 14, 21, 28 and 35 days). An additional control treatment corresponded to plants without biostimulant application. The fruits were evaluated for production and physicochemical attributes: average mass, yield, flesh firmness, titratable acidity, soluble solids content, SSC/TA ratio, pH, total soluble sugars, and weight loss. Fertigation is the recommended application method of biostimulants for yellow melon due to its effect on the increase of average mass, yield, flesh firmness, soluble solids content, and total soluble sugars of the fruits in relation to the spraying method.

ABSTRACT: The purpose of this study was to evaluate the contribution of the inoculation of the strain Azospirillum MAY12 to the development of rice plants of the cultivar BRS Esmeralda. A completely randomized experiment was performed in a greenhouse, with 8 treatments and 6 repetitions, namely, commercial inoculant (CI) containing strains of Azospirillum brasilense ABV5 and ABV6 (T1), MAY12 (T2), nitrogen control with 50 mg of N or full dose (T3), CI + 30% of N (percentage referring to full dose) (T4), CI + 60% N (T5), MAY12 + 30% N (T6), MAY12 + 60% N (T7) and absolute control (T8). There was a significant increase in the fresh and dry mass of the shoot and roots, height, root volume, and the number of tillers and leaves of plants with the MAY12 inoculation (with and without N added) compared to T8. Treatments containing MAY12 + N increased the total dry mass by ~920% compared to T8, matching the T3 yield. The inoculation of MAY12 in isolation was as efficient as the application of the full dose of N, resulting in an increase of ~115% in height and 373% in root volume of the plants compared to T8. The MAY12 strain demonstrated good performance in the development of the cultivar, which was comparable to the CI in the variables tested. Therefore, inoculation with the MAY12 strain is promising for the partial reduction of nitrogen fertilizers in upland rice cultivation.


RESUMO: O objetivo deste trabalho foi avaliar a contribuição da inoculação da estirpe de Azospirillum MAY12 no desenvolvimento de plantas de arroz da cultivar BRS Esmeralda. Foi realizado um experimento inteiramente casualizado em casa de vegetação, tendo oito tratamentos e seis repetições: inoculante comercial (IC) contendo as estirpes de Azospirillum brasilense AbV5 e AbV6 (T1), MAY12 (T2), testemunha nitrogenada com 50 mg de N ou dose cheia (T3), IC + 30% de N (percentual referente à dose cheia) (T4), IC + 60% de N (T5), MAY12 + 30% de N (T6), MAY12 + 60% de N (T7) e testemunha absoluta (T8). Houve aumento significativo da massa fresca e seca da parte aérea e de raízes, altura, volume de raízes e do número de perfilhos e de folhas das plantas com a inoculação de MAY12 (com e sem adição de N) em relação a T8. Os tratamentos contendo MAY12 + N aumentaram em torno de 920% a massa seca total em relação a T8, equiparando-se ao rendimento T3. A inoculação de MAY12, isoladamente, foi tão eficiente quanto a aplicação da dose cheia de N para o aumento em torno de 115% da altura e de 373% do volume de raízes das plantas em relação a T8. A estirpe MAY12 apresentou bom desempenho no desenvolvimento da cultivar equiparando-se com o IC nas variáveis testadas. Portanto, a inoculação com a estirpe MAY12 mostra-se promissora para a redução parcial de fertilizantes nitrogenados no cultivo de arroz de terras altas.
